六味地黃湯加味方이 腦血流 力學에 미치는 機轉硏究

Yukmygihwangtang have been used in oriental medicine for many centuries as a prescription for tonifying and nourishing yin and cerebral blood improvement. YukmygihwangTangGamypang(YTG) consists of the following components ; Rehmanniae Radix Preparat(熟地黃), Dioscoreae Rhizoma(山藥), Corni Fructus(山茱萸), Poria(白茯笭), Moutan Cortex(牧丹皮), Alismatis Rhizoma(澤瀉), Plygalae Radix(遠志), Acori Graminei Rhizoma(石菖蒲), Cervi Pantotrichum Cornu(鹿茸). The effects of YTG on the vascular system is not known. The purpose of this Study was to investigate the effect of YTG on the regional cerebral blood flow(rCBF), mean arterial blood pressure(BP) and action mechanism of rats. The results were as follows ; 1. rCBF was increased by YTG in a dose-dependent manner. 2. BP was inhibited by YTG(0.01mg/kg, P<0.05) in rats. 3. Pretreatment with N^ω-nitro-L-argirinine(_L-NNA) and indomethacin accelerated YTG- induced increased rCBF. 4. Pretreatment with methylene blue(YTG 1.0mg/kg, 10.0mg/kg, P<0.05) and propranolol(YTG 0.01mg/kg, 1.0mg/kg, 10.0mg/kg, P<0.05) inhibited YTG-induced increased rCBF. 5. Pretreatment with _L-NNA, methylene blue(YTG 0.01mg/kg, P<0.05) and indomethacin increased YTG-induced decreased BP. 6. Pretreatment with propranolol inhibited YTG-induced decreased BP. These results suggest that YTG causes a diverse response of rCBF and BP. The rCBF and BP are mediated by cyclic GMP(guanylyl cyclase).
