Effects of Honghwa on the Hepatocyte of Mouse Toxicated by Paraquat

This dissertation is to report the effects of Honghwa upon the hepatocyte of mice induced by Paraquat(PQ). For this study, after medicating Honghwa to mice intoxicated by PQ, I observed its effects on the impaired tissues of liver using electron microscopes, and finally I could get several significant results by comparatively observing the superoxide dismutase(SOD) of hepatocyte and the changes of transaminase activity. 1. In observation using electron microscope, the hepatocyte damage was remarkably observed in every control and experimental group injected with PQ. Particularly in the one-hour group, cell damage was the most noticeable. In the five-day group, the hepatocyte damage began to recover and in the seven-day group, the experimental group showed the aspects of a normal hepatocyte. The count of peroxysomes compared with normal other groups were increased in every group, except seven-day group, particularly in the three-day group. 2. The activity of SOD in anti-oxidation transaminase, MnSOD was the control group reach the peak in the five-day group and reduced in the seven-day group, while, the experimental group reach the peak in the three-day group and grew to reduce from the five-day group. The activity of MnSOD reached the peak in the three-day experimental group compare with any another group. The activity of CuZnSOD began to grow after injection PQ and it reached the peak in the one-day and three-day group, after these it grew to reduce. Compare with the control group, the activity of CuZnSOD was to be high by the one-day experimental group, but reversably it was to be high from the three-day control group. 3. The activity of glutamate-oxaloacetate transaminase(GOT) reach the peak in the five-day group injected with PQ. In contrast to the control group, the activity of GOT in the experimental group was to be low. The activity of glutamate pyruvate transaminase(GPT) also reach the peak in the five-day group and grew to reduce from the seven-day group. The peak of activity of GPT is the five-day control group(113.48±20.38). In conclusion, medicating Honghwa to mice intoxicated by Paraquat is highly effective in anti-oxidation and recovery of hepatocyte but it is necessary to study about long-termed medicating.
