Object: Prevalence of Cerebrovascular accident (CVA) is increasing with the development of economics, the advances in health care and the change of lifestyle. Because CVA has a great influence not on the quality of life, but also on socioeconomic loss, many studies have been conducted about treatment and prevention of this disease. Sokmyeungtang (SMT) has been used for treatment of CVA in oriental traditional medicine, but little has been studied about the effect of protection on the brain cell damage. So this study was designed to evaluate the effect of SMT's protection on brain cell damage against the oxidative stress that was affected by CVA, also investigated the effect of motor function improvement and neurotrophy factor in ischemic cerebral damaged rat. Methods: We measured cell viability after administrating SMT, chemicals (paraquat, SNP, rotenone, and H2O2) which cause oxidative stress, and both SMT and chemicals. Also we observed the change of activity of superoxide dismutase (SOD) and glutathione which were known as anti oxidative enzymes. We carried out neurobehavioral evaluation (rotarod test, Beam-walking test, postural reflex test) and observed BDNF (brain-derived neurotrophic factor) expression by injecting SMT into ischemic cerebral damaged rat. Results: Through this study, we observed the following four results. First, brain cell death caused by paraquat, rotenone, and H2O2 significantly decreased with the treatment of SMT. Second, the activity of SOD and Glutathione slightly decreased with the treatment of SMT. Third, neuronal movement function in ischemic cerebral damaged rat was significantly improved by the treatment of SMT. Forth, BDNF in ischemic cerebral damaged rat increased with the treatment of SMT. In conclusion, SMT protects brain cells from damage induced by oxidative stress(paraquat, rotenone, H2O2). Also SMT improves neuronal movement function and increases BDNF in ischemic cerebral damaged rat.
