Academic researches or documents of rough diamond are limitative because domestic studies on diamond have been focused on loose stones. This research chose 100 pieces of rough diamond from Mobilong Mine, Cameroon. The result of analysis about the crystal habit indicated that octahedral 12%, dodecahedral 24%, trisoctahedral 37%, macle 10% and flat 17%. As the result of analysis about UV fluorescence of rough diamonds, none 44%, faint blue 14%, moderate blue 11%, strong blue 15%, very strong blue 8% and greenish yellow 8% were sorted. According to the optical microscopic observations, characteristic trigons, crystals and feathers were observed and honeycomb structured green strains were found on the surface of natural green diamond. Protogenetic or syngenetic inclusion were found with Micro Raman analysis. Posterite, enstatite, pyrope, coesite, chromite, hematite, goethite and graphite were examined as crystal inclusions. According to the analyses about FT-IR and UV-Vis, Type IaA 23%, IaAB 74% and IaB 3% were shown. Samples with strong UV fluorescence were mostly IaAB. H1a defect center absorption spectrum of natural irradiated green diamond is differentiated by the result that intensity and sharpness is lower than artificial green diamond. As a result of PL analysis, artificial green diamond has higher 3H defect center peak, compared to natural green diamond, which has higher H3 defect center peak. The green colors shown on the surface of rough diamond from Mobilong Mine were caused by natural radioactivity, resulting from the existence of GR1 center.
