ABSTRACT Experimental Studies of extracts and fractions of Phaseolus radiatus L. on the Physiological activity of skin Lee Yu Lim Dept. of Korean Medicine Graduate School of Dongshin University (Directed by Prof. Choi Jeong hwa, K.M.D., Ph. D.) This study is designed to clarify Phaseolus radiatus Extracts and fraction's pharmacological activity on the skin. In this experiment, we were intended to reveal antioxidant, whitening, anti-inflammatory and antibacterial effects of the fractions extracted from the mixture of Phaseolus radiatus L. and ethanol. Polyphenol, flavonoid content, DPPH radical scavenging ability and NO scavenging activity were investigated to confirm the antioxidant activity, and the whitening activity was confirmed by UV blocking activity, tyrosinase inhibiting activity, and melanin formation inhibiting activity. Anti-inflammatory activity is confirmed by measurement of cytotoxicity level by MTT assay and measurement of Cytokine expression, which is the main mediator of inflammation reaction, and antimicrobial activity is confirmed against Staphylococcus aereus and Staphylococcus epidermidis. As a result, overall activity was high in the ethyl acetate fraction. At the concentration of 1000mg / ㎖, the contents of polyphenol 28.01 mg / g and flavonoid 33.40mg / g were measured and 41.12 ± 0.71% and 61.44% of DPPH radical scavenging ability and NO scavenging ability were measured respectively. Tyrosinase inhibitory activity was less than 20% at all concentrations, but the activity to inhibit melanin self-production was higher than that of EF fraction at 32.19 ± 2.79% at 100 μg / ㎖. And Ethyl acetate fraction had a relatively high UV blocking activity . In the anti-inflammatory test, the concentration-dependent activity was shown, and the choloroform and ethyl acetate fractions showed significant NO production inhibitory activity. Cytokine expression was superior to that of the final stage of B cell differentiation, and cell viability was over 80% except for the choloroform fraction at the concentration of 200 μg / ㎖. The antimicrobial activity of Staphylococcus aureus and Staphylococcus epidermidis did not show antimicrobial activity. The results of this experiment confirmed the whitening effect and anti - inflammatory effect of Phaseolus radiatus L.'s extracts and fractions and report the possibility of application as external medicine.
